TheTCRactivatorpresentatthesurfaceofPD-L1/TCRActivatorCHOcellsstimulateTCRinJurkatTcells,whereasoverexpressionofPD-L1ontheCHOcelllineengagesJurkatPD-1,blockingTCRactivationsignalingandpreventingactivationofNFAT.Additionofaneutralizinganti-PD-1oranti-PD-L1antibodytotheco-culturereleasesthePD-L1/PD-1complexandresultsinTCRactivationandincreasedNFATactivity,whichtranslatesintoincreasedluciferasereportersignal.
